    Summary

    Pregnancy reduces sensitivity to angiotensin II. This study found fewer and less-avid angiotensin II receptors in the myometrium of pregnant sheep, explaining this decreased pressor response.

    Background:

    • The pressor response to angiotensin II is diminished in human and ovine pregnancy.
    • The underlying mechanism for this reduced target organ sensitivity remains unclear.

    Purpose of the Study:

    • To investigate pregnancy-induced alterations in angiotensin II receptor characteristics within the myometrium.
    • To determine if changes in receptor number and affinity contribute to decreased angiotensin II sensitivity during gestation.

    Main Methods:

    • Myometrial tissues were collected from nonpregnant and pregnant sheep.
    • Specific binding of radiolabeled angiotensin II (125I-angiotensin II) was quantified in myometrial particulate fractions.
    • Receptor binding assays were performed to assess binding capacity and affinity (Kd).

    • Myometrial angiotensin II binding capacity was significantly lower in pregnant sheep (184 fmol/mg protein) compared to nonpregnant sheep (502 fmol/mg protein).
    • Receptor affinity for angiotensin II was also reduced during pregnancy (Kd 1.72 nM) versus the nonpregnant state (Kd 3.9 nM).

    Conclusions:

    • The data suggest a significant decrease in both the number and affinity of myometrial angiotensin II receptors during pregnancy.
    • These receptor changes likely underlie the reduced pressor responsiveness to angiotensin II observed in pregnant mammals.
