Summary

Small molecules targeting YEATS domains, which bind to acylated lysines in chromatin, are emerging tools. These chemical probes help researchers understand the function of YEATS domains in gene regulation.

Area of Science:

  • Molecular Biology
  • Epigenetics
  • Chemical Biology

Background:

  • Chromatin reader domains recognize histone post-translational modifications.
  • YEATS domains are a recently discovered family of reader domains that bind acylated lysines.
  • Four human proteins (ENL, AF9, YEATS2, YEATS4) contain YEATS domains involved in chromatin and transcription regulation.

Purpose of the Study:

  • To review recent advancements in developing small-molecule tools for YEATS domains.
  • To highlight the utility of these chemical tools in advancing biological understanding of YEATS domain function.

Main Methods:

  • Development of small-molecule probes targeting YEATS domains.
  • Application of these chemical tools to perturb YEATS domain function in biological systems.
