Area of Science:

  • Biomedical Imaging
  • Musculoskeletal Research
  • Radiology

Background:

  • Micro-computed tomography (micro-CT) is a 3D imaging technique limited to dense tissues like bone.
  • Tendons are poorly visualized with micro-CT due to low X-ray absorption.
  • Systematic evaluation of contrast enhancement for tendons across different types and ages is lacking.

Purpose of the Study:

  • To investigate the impact of phosphotungstic acid (PTA) staining and tissue hydration on micro-CT contrast of rat tendons.
  • To establish a method for enhanced 3D visualization of tendons using micro-CT.
  • To explore tendon fascicle organization in situ.

Main Methods:

  • Utilized phosphotungstic acid (PTA) staining to enhance X-ray absorption in rat tendons.
  • Examined tendons from immature (postnatal day 21) and mature (postnatal day 50) rats.
  • Assessed the influence of tissue hydration status on contrast post-staining.
  • Acquired 3D micro-CT images of tail and hindlimb tendons.

Main Results:

  • PTA staining significantly increased tendon X-ray absorption, enhancing contrast for micro-CT imaging.
  • Tissue hydration state critically affected soft tissue contrast after PTA staining.
  • Achieved high-resolution 3D micro-CT visualization of tendons in immature and mature rats.
  • Observed potential fascicle crossing between bundles in tail tendons.

Conclusions:

  • A simple PTA staining and hydration protocol effectively enhances soft tissue contrast for micro-CT visualization of tendons.
  • 3D in situ micro-CT imaging advances the understanding of tendon structure and bone-soft tissue relationships.
  • This technique holds promise for detailed analysis of musculoskeletal tissues.
