    Background:

    • Serum stimulation initiates cell cycle progression in growth-arrested fibroblasts.
    • DNA replication requires the coordinated action of multiple enzymes, including DNA polymerase and primase.

    Purpose of the Study:

    • To investigate the association and activity of DNA polymerase alpha and DNA primase during the cell cycle.
    • To determine the timing of DNA primase activity increase relative to DNA replication.

    Main Methods:

    • Preparation of protein extracts from serum-stimulated mouse 3T3 fibroblasts at various time points.
    • Fractionation of extracts using sucrose gradient centrifugation.
    • Assay of DNA polymerase alpha and DNA primase activities in different fractions.

    Main Results:

    • DNA polymerase alpha and DNA primase exist as a tightly associated complex (8.2-S peak).
    • Neither enzyme was detected independently of the other.
    • Enzyme activities increased concurrently with the onset of the DNA replication phase.
    • DNA primase activity was 5-6 times higher in replicating cells compared to resting cells.
    • A distinct 12.5-S priming activity, likely RNA polymerase I, was also identified.

    Conclusions:

    • DNA polymerase alpha and DNA primase function as a coupled enzymatic unit during cell proliferation.
    • The coordinated increase in activity of both enzymes is essential for initiating DNA replication.
    • RNA polymerase I may play a role in cellular priming activities beyond transcription.
