A novel manganese(II) probe (FX2-MnCl2) enables sensitive detection of adenosine triphosphate (ATP) in lysosomes using multiphoton fluorescence imaging. This advancement allows for monitoring lysosomal ATP migration in vitro for potential clinical diagnosis.
Area of Science:
- Biochemistry
- Chemical Biology
- Molecular Imaging
Background:
- Lysosomal adenosine triphosphate (ATP) levels are crucial for cellular processes.
- Existing methods for monitoring lysosomal ATP face limitations in targeting and resolution.
- Developing specific probes for in situ ATP detection in lysosomes is essential.
Purpose of the Study:
- To develop a novel probe for sensitive and specific in situ monitoring of lysosomal ATP.
- To investigate the probe's properties, including its targeting and fluorescence enhancement capabilities.
- To demonstrate the probe's utility in visualizing lysosomal ATP dynamics.
Main Methods:
- Synthesis and characterization of a manganese(II) complex (FX2-MnCl2) with near-infrared nonlinear optical properties.
- Utilizing multiphoton fluorescence imaging (MP-FI) and multiphoton fluorescence lifetime imaging microscopy (MP-FLIM).
- Employing dual recognition sites (diethoxy and manganese ions) for ATP binding.
Main Results:
- The FX2-MnCl2 probe exhibits a fivefold fluorescence enhancement in the presence of ATP.
- The probe demonstrates specific targeting to lysosomes.
- Lysosome-specific ATP migration was successfully monitored under near-infrared laser excitation.
- The probe exhibits a slight fluorescence lifetime change upon ATP binding.
Conclusions:
- FX2-MnCl2 is a novel multiphoton fluorescent complex for lysosomal ATP detection.
- The probe offers enhanced sensitivity and specificity compared to existing methods.
- This technology holds potential as a biomarker for lysosomal ATP in in vitro diagnostics.
