The transcription factor Bhlhe40 promotes T helper 1 (Th1) cell differentiation and inhibits T follicular helper (Tfh) cell development. Deleting Bhlhe40 in CD4+ T cells enhances Tfh cell differentiation and antibody production.
Area of Science:
- Immunology
- Molecular Biology
- Transcriptional Regulation
Background:
- Naive CD4+ T cells differentiate into T helper 1 (Th1) or T follicular helper (Tfh) cells, crucial for cellular and humoral immunity, respectively.
- While T-bet and BCL-6 are known drivers of Th1 and Tfh lineages, additional transcriptional regulators governing this fate bifurcation remain incompletely understood.
Purpose of the Study:
- To investigate the role of the transcriptional regulator Bhlhe40 in dictating the Th1/Tfh differentiation axis in mice.
- To elucidate the molecular mechanisms by which Bhlhe40 influences Th1 versus Tfh lineage commitment.
Main Methods:
- CD4+ T cell-specific deletion of Bhlhe40 in mice.
- Epigenetic profiling of virus-specific Th1 and Tfh cells following LCMV infection.
- Analysis of gene expression and protein binding sites.
Main Results:
- CD4+ T cell-specific deletion of Bhlhe40 abrogated Th1 differentiation while augmenting Tfh differentiation.
- Deletion of Bhlhe40 led to increased germinal center B cells and antibody production.
- Bhlhe40 was found to directly bind cis-regulatory elements, activating Th1-related genes (e.g., Tbx21, Cxcr6) and repressing Tfh-related genes (e.g., Bcl6, Cxcr5).
Conclusions:
- Bhlhe40 acts as a critical transcription factor that promotes Th1 cell differentiation.
- Bhlhe40 functions as a repressor to suppress Tfh cell differentiation.
- Bhlhe40 influences not only Tfh cell differentiation but also their capacity to support B cell responses.

NK cells are a crucial part of our innate immune system, acting as the first line of defense against viral infections. These cells can recognize and kill infected cells without prior exposure to the virus, effectively slowing down the spread of infection. Additionally, NK cells produce proinflammatory...
