This study developed a novel drug delivery system for clindamycin (CLY) to treat diabetic foot infections (DFI). The bacteria-sensitive microparticle patch (MP-CLY-SEMAP) improved drug delivery and reduced bacterial viability.
Area of Science:
- Biomaterials Science
- Drug Delivery Systems
- Dermatology
Background:
- Diabetic foot infection (DFI) is a common complication of diabetes mellitus.
- Current treatments with clindamycin (CLY) via oral or intravenous routes cause significant side effects.
- There is a need for improved localized drug delivery methods for DFI.
Purpose of the Study:
- To develop and characterize a novel bacteria-sensitive microparticle system (MP-CLY) for clindamycin (CLY) encapsulation.
- To formulate MP-CLY into a separable effervescent microarray patch (MP-CLY-SEMAP) for enhanced skin penetration and localized delivery.
- To evaluate the dermatokinetic profile and antibacterial efficacy of the MP-CLY-SEMAP system.
Main Methods:
- Clindamycin (CLY) was encapsulated into polycaprolactone (PCL) microparticles (MP-CLY).
- MP-CLY was incorporated into a separable effervescent microarray patch (MP-CLY-SEMAP).
- Characterization included encapsulation efficiency, in vitro release studies, mechanical properties, and ex vivo dermatokinetic testing on infected skin models.
Main Results:
- MP-CLY showed high encapsulation of CLY with no chemical structure alteration.
- In vitro release demonstrated bacteria-triggered and controlled release of CLY.
- MP-CLY-SEMAP exhibited optimal mechanical properties and efficient skin penetration, leading to a 99.99% reduction in bacterial viability ex vivo.
Conclusions:
- The MP-CLY-SEMAP system represents a proof-of-concept for improved localized delivery of CLY for DFI.
- This novel delivery system enhances the dermatokinetic profile of CLY.
- Further in vivo studies in animal models are warranted to validate the efficacy and safety of MP-CLY-SEMAP.
