This study developed aptamer-functionalized magnetic microparticles to capture Streptococcus pneumoniae (S. pneumoniae). This novel biosensor assay effectively isolates bacteria from blood and PBS samples, improving signal-to-noise ratio.
Area of Science:
- Biotechnology
- Biosensor Development
- Molecular Diagnostics
Background:
- Biosensor advancement is limited by signal-to-noise ratio issues.
- Interfering factors like blood in samples reduce biosensor accuracy.
- Selective aptamer binding offers a solution to target specific analytes.
Purpose of the Study:
- To develop a novel assay for isolating Streptococcus pneumoniae (S. pneumoniae).
- To utilize aptamer-decorated magnetic microparticles for bacterial capture.
- To overcome sample matrix interference in biosensing.
Main Methods:
- Selected aptamers with high affinity for S. pneumoniae and no affinity for non-target cells.
- Decorated magnetic microparticles with these specific aptamers.
- Developed a capture assay to isolate S. pneumoniae from PBS and blood.
Main Results:
- Achieved high capture efficiencies for S. pneumoniae.
- Demonstrated successful isolation from both phosphate-buffered saline (PBS) and blood matrices.
- Reported capture efficiencies of up to 87% in PBS and 66% in blood samples.
Conclusions:
- Aptamer-functionalized magnetic microparticles provide an effective method for S. pneumoniae isolation.
- The developed assay demonstrates high specificity and efficiency in complex sample matrices.
- This approach enhances biosensor performance by reducing interference.
