    Background:

    • Natural killer (NK) cells are crucial for innate immunity against cancer.
    • Spaceflight, including microgravity, may compromise immune defenses, increasing cancer risk for astronauts.

    Approach:

    • Human NK cells were cultured under normal and simulated microgravity (sµG) conditions.
    • NK cell cytotoxicity against leukemia cell lines (K-562 and MOLT-4) was assessed.
    • RNA sequencing was performed to investigate the molecular mechanisms of sµG effects.

    Key Points:

    • Even brief exposure to sµG markedly reduced NK cell cytotoxicity against both CML and T-ALL cell lines.
    • Continuous sµG exposure exacerbated the reduction in NK cell function.
    • RNA-seq data provided insights into the mechanisms underlying reduced NK cell cytotoxicity.

    Conclusions:

    • Microgravity impairs NK cell-mediated anti-leukemia defense.
    • This impairment may increase astronaut susceptibility to leukemogenesis during extended space missions.
    • Further research is needed to understand and mitigate these risks.
