Background:

  • Seminal cryopreservation can damage sperm, necessitating optimized processing methods.

Purpose of the Study:

  • To compare the efficacy of density gradient processing versus washing/centrifugation for seminal plasma removal prior to cryopreservation.
  • To assess the impact of these methods on key semen parameters after thawing.

Main Methods:

  • Semen samples from 26 normozoospermic patients were divided into three groups: with seminal plasma, after washing/centrifugation, and after density gradient processing.
  • All samples were cryopreserved for at least two weeks.
  • Sperm motility, count, morphology, and viability were evaluated before cryopreservation and after thawing.

Main Results:

  • Density gradient processing significantly improved the selection of motile, viable sperm with normal morphology in fresh samples (p<0.05).
  • Cryopreservation negatively impacted all semen parameters irrespective of the processing method.
  • Despite lower sperm recovery post-thaw, density gradient processing resulted in significantly higher progressive motility, total motility, viability, and morphology (p<0.05).

Conclusions:

  • Cryopreservation significantly compromises sperm motility, morphology, and viability.
  • Density gradient processing is superior in selecting higher-quality spermatozoa compared to other methods in normozoospermic individuals.
  • The benefits of density gradient processing for sperm quality are maintained even after cryopreservation and thawing.
