Enzyme-Linked Immunosorbent Assay01:33

Enzyme-Linked Immunosorbent Assay

12.9K
In 1971, Peter Perlman and Eva Engvall developed an Enzyme-linked immunosorbent assay (ELISA or EIA). ELISA differs from western blot in that the assays are conducted in microtiter plates or in vivo rather than on an absorbent membrane.
There are many different types of ELISAs, but they all involve an antibody molecule whose constant region binds an enzyme, leaving the variable region free to bind its specific antigen.  Enzyme-substrate reaction allows the antigen to be visualized or...

Area of Science:

  • Biomedical Engineering
  • Nanotechnology
  • Parasitology

Background:

  • Leishmaniasis diagnosis requires sensitive and specific methods.
  • The surface protease (Gp63) of Leishmania major is a key diagnostic target.
  • Gold nanoparticles (AuNPs) offer unique optical properties for biosensing.

Purpose of the Study:

  • To develop a novel colorimetric biosensor for detecting antibodies against Leishmania major surface protease (Gp63).
  • To utilize gold nanoparticles (AuNPs) as a colorimetric indicator for antibody detection.
  • To establish the sensitivity and reliability of the developed biosensor for leishmaniasis diagnosis.

Main Methods:

  • Conjugation of kinetoplastid membrane protein-11 (KMP-11) to gold nanoparticles (AuNPs).
  • Colorimetric detection of anti-Gp63 antibodies via AuNP aggregation/dispersion.
  • UV-vis spectroscopy and visual color change analysis for detection.

Main Results:

  • The biosensor detected anti-Gp63 antibodies at dilutions up to 1:640.
  • A distinct color change from red/pink to purple/violet indicated positive results.
  • High recovery rates (95.3%–103.1%) and low relative standard deviation (1.29%) were achieved.

Conclusions:

  • The developed gold nanoparticle-based colorimetric biosensor is a sensitive and reliable tool for detecting anti-Gp63 antibodies.
  • This assay shows potential for rapid and accessible diagnosis of leishmaniasis.
  • The biosensor demonstrated effective detection in crude antigen and serum samples.
