Nicotine promotes skin cutaneous melanoma (SKCM) progression by activating the α5-nicotinic acetylcholine receptor (α5-nAChR) and ADAM10 signaling pathway. This pathway enhances cancer stemness and migration, leading to poorer patient survival.
Area of Science:
- Oncology
- Dermatology
- Molecular Biology
Background:
- Skin cutaneous melanoma (SKCM) is a deadly skin cancer with increasing incidence.
- Smoking is a known risk factor and poor prognostic marker for melanoma.
- Nicotine, a component of smoke, influences SKCM cell behavior through receptors like α5-nicotinic acetylcholine receptor (α5-nAChR).
Purpose of the Study:
- To investigate the relationship between α5-nicotinic acetylcholine receptor (α5-nAChR) and ADAM10 in nicotine-associated SKCM.
- To elucidate the role of the α5-nAChR/ADAM10 signaling axis in melanoma progression.
Main Methods:
- Correlation analysis of α5-nAChR and ADAM10 expression in SKCM patient data.
- Investigating the mechanism of nicotine-induced ADAM10 expression via STAT3.
- Assessing the impact of the α5-nAChR/ADAM10 axis on SKCM cell stemness and migration.
- Analyzing marker expression in nicotine-treated mouse homograft models.
Main Results:
- α5-nAChR expression positively correlated with ADAM10 expression and reduced SKCM patient survival.
- α5-nAChR mediated nicotine-induced ADAM10 expression through the STAT3 pathway.
- The α5-nAChR/ADAM10 signaling pathway was implicated in SKCM cell stemness and migration.
- Expression of ADAM10, EMT markers, and stemness markers was associated with α5-nAChR in nicotine-exposed mouse models.
Conclusions:
- The α5-nicotinic acetylcholine receptor (α5-nAChR)/ADAM10 signaling pathway plays a crucial role in nicotine-induced melanoma progression.
- Targeting this pathway may offer therapeutic strategies for smokers diagnosed with SKCM.
