GABAergic scaling, not AMPAergic scaling, may be the primary mechanism for controlling neuron firing rates. This study demonstrates GABAergic scaling adjusts to maintain stable spiking activity in cortical cultures.
Area of Science:
- Neuroscience
- Cellular Neuroscience
- Synaptic Plasticity
Background:
- Homeostatic plasticity mechanisms aim to stabilize neural function.
- AMPAergic scaling was previously considered a key regulator of neuronal spiking activity.
- Recent evidence questions the direct role of AMPAergic scaling in response to spiking changes.
Purpose of the Study:
- To investigate the role of GABAergic scaling in homeostatic control of neuronal spiking.
- To compare the efficacy of GABAergic versus AMPAergic scaling in regulating spike rates.
- To determine how changes in AMPA receptor (AMPAR) and GABAR transmission affect homeostatic plasticity.
Main Methods:
- Perturbations were applied to cortical cultures to alter spiking activity.
- Changes in GABAergic synaptic transmission were measured in response to spiking alterations.
- The influence of AMPAR and GABAR transmission levels on GABAergic scaling was examined.
Main Results:
- Increased spiking led to multiplicative GABAergic upscaling.
- Decreased spiking resulted in multiplicative GABAergic downscaling.
- Changes in AMPAR or GABAR transmission indirectly affected GABAergic scaling via spiking activity.
Conclusions:
- GABAergic scaling actively controls neuronal spiking levels in cortical cultures.
- GABAergic scaling appears to be a more significant homeostatic mechanism for spike rate regulation than AMPAergic scaling.
- Spiking activity is the primary driver for GABAergic scaling, rather than direct modulation of synaptic transmission.
