In 1971, Peter Perlman and Eva Engvall developed an Enzyme-linked immunosorbent assay (ELISA or EIA). ELISA differs from western blot in that the assays are conducted in microtiter plates or in vivo rather than on an absorbent membrane.

  • To evaluate the Immunoturbidimetry assay against the Siemens BNII® Nephelometry method routinely used in the clinical laboratory.

Main Methods:

  • Analytical performance comparison of Immunoturbidimetry (TBS Optilite®) and Nephelometry (Siemens BNII®) for IgG and IgG subclass quantification.
  • Evaluation of assay reliability and correlation between the sum of IgG subclasses and Total IgG.

Main Results:

  • The Immunoturbidimetry assay demonstrated reliability in evaluating IgG subclasses.
  • The sum of IgG subclasses and Total IgG showed better correlation using the Immunoturbidimetry assay compared to Nephelometry.
  • Despite sharing a similar principle, direct comparison of results between the two methodologies was compromised.

Conclusions:

  • Immunoturbidimetry appears to be a reliable method for IgG subclass evaluation.
  • Further studies are needed to determine the optimal methodology for specific patient populations before switching assays.
  • Standardization of IgG subclass assays is essential to reduce discrepancies and improve diagnostic accuracy.
