Lateral olfactory tract usher substance (LOTUS) promotes neurite outgrowth by interacting with Teneurin-4 (Ten-4). This interaction is independent of Nogo receptor-1 (NgR1) and suggests a new mechanism for neuronal development and CNS repair.
Area of Science:
- Neuroscience
- Molecular Biology
- Cell Biology
Background:
- Neurite outgrowth is vital for neuronal circuit formation and CNS regeneration.
- Axonal growth inhibitors like Nogo/NgR1 impede regeneration in the adult mammalian CNS.
- Lateral olfactory tract usher substance (LOTUS) previously showed potential as an antagonist for NgR1.
Purpose of the Study:
- To investigate the molecular mechanisms underlying LOTUS-mediated neurite outgrowth.
- To identify novel LOTUS-binding proteins involved in neuronal development and regeneration.
Main Methods:
- Neurite outgrowth assays using cultured retinal explant neurons on LOTUS-coated substrates.
- Screening for LOTUS-binding proteins using liquid chromatography-tandem mass spectrometry (LC-MS/MS).
- Validation of interactions using pull-down assays and RNA interference (RNAi) knockdown of Teneurin-4 (Ten-4).
Main Results:
- LOTUS promotes neurite outgrowth independently of NgR1.
- LOTUS directly interacts with Teneurin-4 (Ten-4), a cell adhesion molecule.
- Knockdown of Ten-4 or addition of soluble Ten-4 inhibits LOTUS-mediated neurite outgrowth.
Conclusions:
- LOTUS promotes neurite outgrowth through interaction with Teneurin-4 (Ten-4).
- This LOTUS-Ten-4 pathway offers a new molecular mechanism for neuronal circuit formation and CNS injury repair.
