Human umbilical cord mesenchymal stem cell-derived exosomes (HucMSCs-EXOs) loaded with anti-programmed cell death 1 (PD-1) peptide, named MEP, effectively treat septic-induced acute lung injury (ALI). MEP outperforms individual components in reducing inflammation and improving lung injury in mice.
Area of Science:
- Biomedical Engineering
- Immunology
- Regenerative Medicine
Background:
- Sepsis-induced cytokine storm can cause acute lung injury (ALI).
- Human umbilical cord mesenchymal stem cell-derived exosomes (HucMSCs-EXOs) exhibit immunosuppressive and anti-inflammatory properties.
- Programmed cell death receptor 1 (PD-1) is vital for immune homeostasis.
Purpose of the Study:
- To investigate the synergistic therapeutic effect of exosomes loaded with anti-PD-1 peptide (MEP) on septic-ALI.
- To elucidate the therapeutic mechanism of MEP in treating septic-ALI.
Main Methods:
- Engineering HucMSCs-EXOs to create MEP by loading anti-PD-1 peptide.
- Evaluating MEP's therapeutic effect in vivo (septic-ALI mice) and in vitro.
- Analyzing inflammatory markers, cytokine levels, cell apoptosis, macrophage polarization, and gene expression.
Main Results:
- MEP significantly improved lung injury index in septic-ALI mice compared to individual components.
- MEP reduced inflammatory markers (CRP, PCT) and pro-inflammatory cytokines (TNF-α, IL-1β).
- MEP increased anti-inflammatory cytokine (IL-10), CD68+ macrophages, promoted M2 macrophage polarization, and decreased inflammasome-related genes.
Conclusions:
- MEP demonstrates superior therapeutic efficacy for septic-ALI compared to anti-PD-1 peptide or EXOs alone.
- MEP effectively modulates immune responses and reduces inflammation in septic-ALI.
- MEP shows promise as a novel therapeutic agent for treating septic-ALI.
