Ribosome biogenesis (RiBi) is essential for generating hematopoietic stem cells (HSCs) from hemogenic endothelial cells (HECs). Inhibiting RiBi blocks HSC generation, highlighting its critical role in this process.
Area of Science:
- Developmental Biology
- Hematopoiesis
- Cell Biology
Background:
- Hemogenic endothelial cells (HECs) arise from embryonic aortic endothelial cells.
- HECs generate hematopoietic stem cells (HSCs).
- Ribosome biogenesis (RiBi) is upregulated in HSC-primed HECs.
Purpose of the Study:
- To determine if RiBi is required for HSC generation from HECs.
- To investigate the role of RiBi in the endothelial-to-hematopoietic transition.
Main Methods:
- Pharmacological inhibition of RiBi in mouse explant cultures.
- Disruption of RiBi in HECs and T1 pre-HSCs.
- Analysis of Runx1 binding to RiBi gene loci.
Main Results:
- Robust RiBi is augmented during endothelial-to-hematopoietic transition in mice.
- Pharmacological RiBi inhibition completely blocked HSC generation.
- Disrupting RiBi selectively impaired HSC generation potential of HECs.
- Runx1 directly binds RiBi gene loci upon HEC specification.
Conclusions:
- RiBi is indispensable for generating HSCs from HECs.
- Findings provide insights for improving HSC regeneration strategies.
- RiBi plays a critical role in regulating cell cycle activity during HSC generation.
