Overview Of Cell Separation And Isolation01:20

Overview Of Cell Separation And Isolation

5.6K
Cell separation was first achieved in 1964 by S. H. Seal, who separated large tumor cells from the smaller blood cells using filtration. Two years later, Pohl and Hawk performed experiments on how cells respond differently to a nonuniform electric field based on the cell type. Such observations were the inception of cell separation methods, which allow isolating a single cell type from a heterogeneous sample.

Subcellular Fractionation01:32

Subcellular Fractionation

6.9K
The homogenate obtained after cell lysis contains various membrane-bound organelles that can be further separated into pure fractions by subcellular fractionation. These isolates are used to study specific cellular components, analyze localized protein activity, and are even employed in diagnostics. Fractionation is typically achieved using centrifugation methods, the most common being density-gradient and differential centrifugation.

    • Biotechnology
    • Cell Biology
    • Microfluidics

    Background:

    • Glycolysis is a key metabolic pathway linked to cell state and disease.
    • Current cell sorting methods often require labels or lack precision in metabolic profiling.

    Purpose of the Study:

    • To present a passive, label-free droplet microfluidic platform for stepwise cell sorting based on glycolysis.
    • To expand the Sorting by Interfacial Tension (SIFT) technique for multi-level metabolic cell isolation.

    Main Methods:

    • Utilized a microfluidic device employing Sorting by Interfacial Tension (SIFT).
    • Exploited changes in droplet interfacial tension driven by pH variations from cellular glycolysis and surfactant adsorption.
    • Developed a system with multiple microfabricated rails for stepwise cell isolation.

    Main Results:

    • Successfully sorted cells into three distinct populations based on glycolytic activity.
    • Correlated sorted cell populations with a surface marker for T-cell activation.
    • Demonstrated the platform's capability for label-free, high-throughput metabolic cell sorting.

    Conclusions:

    • The developed SIFT platform offers a novel approach for precise, label-free cell sorting based on metabolic function.
    • This technology has significant potential for advancing research in oncology, immunology, and immunotherapy by enabling detailed analysis of critical cell subpopulations.
    • Facilitates the study of cell states and diseases associated with altered glycolytic metabolism.
