Biofilms

1
Biofilms are complex communities of microorganisms encased in a self-produced extracellular polysaccharide matrix attached to surfaces. These microbial consortia can include single or multiple species, providing enhanced survival benefits by forming organized, multilayered structures. The formation of biofilms occurs through four key stages: attachment, colonization, development, and dispersal.During attachment, free-swimming planktonic cells adhere to a surface, often facilitated by...

Area of Science:

  • Phytochemistry
  • Microbiology
  • Antimicrobial Agents

Background:

  • Cryptocarya species extracts exhibit antimicrobial properties.
  • Fractionation of extracts can enhance their antimicrobial potential.
  • Biofilm formation by Candida albicans and Streptococcus mutans poses significant health challenges.

Purpose of the Study:

  • To evaluate the antimicrobial activity of Cryptocarya moschata fractions against planktonic cells and biofilms.
  • To determine the efficacy of different fractions against Candida albicans and Streptococcus mutans.

Main Methods:

  • Four fractions of C. moschata were prepared: hexane, acetate/hexane (1:1), ethyl acetate, and water.
  • Antimicrobial activity was assessed using colony-forming unit counts (CFU/mL) for planktonic cells and biofilms.
  • Biofilm inhibition was further evaluated by measuring cell metabolic activity and using confocal laser scanning microscopy (CLSM).

Main Results:

  • Acetate/hexane (1:1) and 100% ethyl acetate fractions reduced planktonic Candida albicans.
  • No significant effect was observed on planktonic Streptococcus mutans.
  • All tested C. moschata fractions reduced biofilm CFU/mL by 2-5 logs and cell metabolic activity by approximately 80%.
  • CLSM revealed reduced microorganism viability and extracellular matrix damage in biofilms.

Conclusions:

  • Cryptocarya moschata fractions, particularly acetate/hexane (1:1) and 100% ethyl acetate, demonstrate significant antimicrobial activity against biofilms.
  • These fractions hold potential for developing novel anti-biofilm agents.
