Electrophilic Aromatic Substitution: Fluorination and Iodination of Benzene01:13

Electrophilic Aromatic Substitution: Fluorination and Iodination of Benzene

5.8K
Bromination and chlorination of aromatic rings by electrophilic aromatic substitution reactions are easily achieved, but fluorination and iodination are difficult to achieve. Fluorine is so reactive that its reaction with benzene is difficult to control, resulting in poor yields of monofluoroaromatic products. To address this, Selectfluor reagent is used as a fluorine source in which a fluorine atom is bonded to a positively charged nitrogen.

Targeted Cancer Therapies

7.5K
The targeted cancer therapies, also known as “molecular targeted therapies,” take advantage of the molecular and genetic differences between the cancer cells and the normal cells. It needs a thorough understanding of the cancer cells to develop drugs that can target specific molecular aspects that drive the growth, progression, and spread of cancer cells without affecting the growth and survival of other normal cells in the body.
There are several types of targeted therapies against...

Area of Science:

  • Coordination Chemistry
  • Medicinal Chemistry
  • Materials Science

Background:

  • N-heterocyclic carbenes (NHCs) are versatile ligands in coordination chemistry.
  • Silver(I) complexes have demonstrated significant biological activities.
  • Developing novel anticancer agents with improved selectivity is a critical need.

Purpose of the Study:

  • Synthesize and characterize novel functionalized (benz)imidazolium NHC ligands and their silver(I) complexes.
  • Evaluate the anticancer activity and selectivity of these compounds against breast cancer cell lines.
  • Investigate the mechanism of action, including cellular uptake and DNA interaction.

Main Methods:

  • Synthesis of five new NHC ligands and four Ag(I)-NHC complexes.
  • Full characterization including single crystal X-ray diffraction.
  • In vitro screening against breast and noncancerous cell lines, ICP-MS for silver uptake, DNA interaction studies, molecular docking, and ROS evaluation.

Main Results:

  • The mononuclear benzimidazole complex exhibited the highest cytotoxicity.
  • The binuclear benzimidazole complex demonstrated superior cancer cell selectivity.
  • A strong correlation was observed between cellular silver uptake and cytotoxicity.
  • The binuclear complex showed promising activity against MDA-MD-231 breast cancer cells.

Conclusions:

  • The synthesized Ag(I)-NHC complexes show potential as anticancer agents.
  • The binuclear benzimidazole complex is a highly selective candidate for treating late-stage breast cancer.
  • Further development could lead to novel therapeutic strategies for breast cancer treatment.
