Area of Science:

  • Immunology
  • Pharmacology
  • Mycology

Background:

  • Lentinula edodes polysaccharides are known immunomodulators.
  • SeLe30 is a novel selenium-enriched glucan mixture derived from L. edodes mycelium.

Purpose of the Study:

  • To evaluate the immunomodulatory properties of SeLe30 on human T cells.
  • To analyze SeLe30's effects on T cell activation markers, subsets, and cytokine production.

Main Methods:

  • Human peripheral blood mononuclear cells (PBMCs) and T cells were isolated.
  • Cells were treated with SeLe30 under different stimulation conditions (unstimulated, anti-CD3 Ab, anti-CD3/CD28 Abs).
  • Flow cytometry and cytokine assays were used to analyze T cell responses.

Main Results:

  • SeLe30 modulated the expression of CD25, CD366, and CD279 on T cells depending on stimulation.
  • SeLe30 altered the proportions of CD8+ T cell subsets (naïve, central memory, effector memory).
  • SeLe30 upregulated cytokine production (IFN-γ, IL-6, TNF-α) and exhibited context-dependent effects on T cell activation.

Conclusions:

  • SeLe30 demonstrates context-dependent immunomodulatory effects on human T cells.
  • It influences T cell activation, immune checkpoint expression, and cytokine profiles.
  • SeLe30's overall impact involves indirect interactions with other immune cells.
