Background:

  • Parathyroid carcinoma (PC) is a rare endocrine malignancy with a poor prognosis.
  • Accurate diagnosis of PC is challenging, necessitating novel diagnostic markers.
  • Aberrant glycosylation is implicated in various cancers, but its role in PC is understudied.

Purpose of the Study:

  • To investigate glycosylation patterns in PC using lectin microarray.
  • To identify potential diagnostic markers for differentiating PC from parathyroid adenoma (PA).

Main Methods:

  • Assessed 8 PC and 6 PA tissues using a 70-lectin microarray.
  • Compared lectin-specific glycosylation patterns between PC and PA groups.
  • Validated differentially expressed lectins using lectin histochemistry.

Main Results:

  • Significant differences in lectin binding were observed in 71.4% of lectins between PC and PA (P < 0.05).
  • Amaranthus caudatus (ACL) lectin showed the most significant differential response, with higher intensity in PC tissues (ratio = 2.45).
  • Histochemistry confirmed elevated ACL intensity in PC compared to PA; differentially expressed glycans were primarily glucose, mannose, and galactose-based.

Conclusions:

  • PC exhibits distinct glycomic features compared to PA.
  • Amaranthus caudatus (ACL) lectin demonstrates potential as a candidate diagnostic marker for parathyroid carcinoma.
