Cancer Survival Analysis01:21

Cancer Survival Analysis

Cancer survival analysis focuses on quantifying and interpreting the time from a key starting point, such as diagnosis or the initiation of treatment, to a specific endpoint, such as remission or death. This analysis provides critical insights into treatment effectiveness and factors that influence patient outcomes, helping to shape clinical decisions and guide prognostic evaluations. A cornerstone of oncology research, survival analysis tackles the challenges of skewed, non-normally...

Summary

High CCL17 chemokine levels in thyroid carcinoma (THCA) correlate with increased immune cell infiltration and poorer patient survival. This suggests CCL17 has prognostic value for tumor immunity in THCA.

Area of Science:

  • Oncology
  • Immunology
  • Bioinformatics

Background:

  • CCL17, a chemokine, is implicated in various cancers by modulating immune cell infiltration into the tumor microenvironment (TME).
  • The role of CCL17 in thyroid carcinoma (THCA) and its impact on tumor immunity remain largely unexplored.

Purpose of the Study:

  • To investigate the association between CCL17 expression levels and its prognostic value in thyroid carcinoma (THCA).
  • To explore the relationship of CCL17 with tumor immunity, including immune cell infiltration and clinical characteristics in THCA.

Main Methods:

  • Bioinformatic analysis using GEPIA, TCGA, Kaplan-Meier plotter, WGCNA, STRING, GO, KEGG, TISIDB, ESTIMATE, CIBERSORT, and TIMER databases.
  • Weighted gene co-expression network analysis (WGCNA) to identify key gene modules and hub genes.
  • Immunohistochemistry validation of CCL17 expression in tumor and para-carcinoma tissues.

Main Results:

  • CCL17 was significantly upregulated in THCA tissues compared to normal samples, with high expression linked to decreased overall survival.
  • High CCL17 expression correlated with advanced tumor stage, T classification, N classification, and higher CCR4 expression.
  • CCL17, identified as a hub gene, was associated with immune cell infiltration, cytokine-cytokine receptor interaction, and chemokine signaling pathways.

Conclusions:

  • CCL17 expression is a significant prognostic biomarker for thyroid carcinoma, associated with immune cell infiltration and adverse patient outcomes.
  • CCL17 may accelerate tumor progression in THCA, potentially by promoting lymph node metastasis and influencing the tumor immune microenvironment.
