Near infrared (NIR)-mediated nicotinamide adenine dinucleotide (NADH) pre-treatment improves isolated mitochondria function. This enhanced mitochondrial therapy shows promise for treating mitochondrial disorders and muscle degeneration.
Area of Science:
- Cell Biology
- Mitochondrial Medicine
- Regenerative Medicine
Background:
- Mitochondrial transfer is a promising cell therapy due to low immunogenicity.
- Isolated mitochondria lose function rapidly outside the cell, limiting therapeutic potential.
- Nicotinamide adenine dinucleotide (NADH) is crucial for mitochondrial energy production but has poor cellular uptake.
Purpose of the Study:
- To investigate if NIR-mediated NADH pre-treatment enhances mitochondrial function and stability.
- To develop a photo-mediated method for improved NADH delivery and absorption into cells.
- To evaluate the efficacy of enhanced mitochondria in treating muscle dysfunction.
Main Methods:
- L6 cells were treated with NADH and irradiated with near-infrared (NIR) light.
- Mitochondrial function, energy production, and stability were assessed.
- Enhanced mitochondria (P-MT) were encapsulated in liposomes and delivered to dysfunctional muscle cells.
Main Results:
- NIR-mediated NADH uptake significantly improved mitochondrial energy production and function in L6 cells.
- Enhanced mitochondrial function was maintained after isolation from donor cells.
- Liposome-encapsulated P-MT promoted greater mitochondrial recovery and muscle regeneration compared to conventional mitochondria.
Conclusions:
- NIR-mediated NADH delivery is an effective strategy to enhance mitochondrial function and stability.
- Pre-treated mitochondria (P-MT) demonstrate improved therapeutic potential for mitochondrial disorders.
- This approach offers a novel treatment strategy for muscle degeneration and related conditions.

Translocation of Proteins into the Mitochondria
Sorting of outer membrane proteins:
Mitochondrial outer membrane proteins are of two types: the transmembrane, beta-barrel porins, and the membrane-anchored, alpha-helical proteins. Beta-barrel porin precursors are translocated by the TOM complex and inserted into the outer mitochondrial membrane by the SAM complex. In contrast,...
