Summary

Psoriasis increases the risk of developing primary Sjögren's syndrome (PSS), but PSS does not increase the risk of psoriasis. This study clarifies the causal link between these two autoimmune conditions.

Background:

  • Primary Sjögren's syndrome (PSS) and psoriasis frequently co-occur.
  • The causal relationship and underlying mechanisms between PSS and psoriasis are not well understood.

Purpose of the Study:

  • To investigate the bidirectional causal relationship between PSS and psoriasis using Mendelian randomization (MR).

Main Methods:

  • Bidirectional MR analysis was performed using four MR methods: Inverse Variance Weighted (IVW), MR-Egger regression, weighted median, and weighted mode.
  • Sensitivity analyses were conducted to assess heterogeneity and pleiotropy, ensuring the robustness of the findings.

Main Results:

  • A significant causal effect of psoriasis on the risk of developing PSS was identified (IVW, P = 0.028).
  • No significant causal effect of PSS on the risk of developing psoriasis was observed (IVW, P = 0.328).

Conclusions:

  • Psoriasis is a causal risk factor for developing primary Sjögren's syndrome.
  • The findings provide novel insights into the etiology of both PSS and psoriasis, highlighting a unidirectional causal link.
