Inborn Errors of Metabolism01:20

Inborn Errors of Metabolism

120
Phenylketonuria (PKU) is a protein metabolism disorder characterized by high blood levels of the amino acid phenylalanine. This results from a mutation in the gene responsible for phenylalanine hydroxylase, an enzyme that converts phenylalanine into tyrosine. When this enzyme is deficient, phenylalanine builds up in the blood, leading to symptoms such as vomiting, rashes, seizures, growth deficiency, and severe mental retardation. An early diagnosis and a diet restricting phenylalanine intake...

Factors Affecting the Risk of Infection01:26

Factors Affecting the Risk of Infection

10.7K
The hosts' susceptibility to infection depends on several factors. The integrity of the skin and mucous membranes helps protect the body against microbial attacks. When the skin is altered, the chance of infection, limb loss, and even death increases.
The integrity and count of the white blood cells help the body resist pathogens and fight infection. When impaired, it reduces the body's resistance to pathogens. The acidic pH levels of the gastrointestinal, genitourinary tracts, and skin...

Cholesterol: Significance and Regulation

483
Although not a source of energy, cholesterol plays a significant role as a foundational structure for bile salts, steroid hormones, and vitamin D, as well as being a crucial component of plasma membranes. Approximately 15% of blood cholesterol is derived from our diet, with the remainder synthesized from acetyl CoA by the liver and intestines. Cholesterol is eliminated from the body through its conversion into bile salts, which are eventually discarded in the feces.
Considering cholesterol and...

Cancer Prevention02:59

Cancer Prevention

6.0K
Several factors can increase the risk of cancer in an individual. About 50% of cancer cases can be prevented by adopting a healthy lifestyle, regular exercise, eating healthy, and following a modest cancer prevention diet. Epidemiological studies have consistently shown that populations with vegetable and fruit-rich diets have reduced the incidence of cancer. On the other hand, populations who have a diet rich in animal fat, red meat, junk food, or high calories are predisposed to cancer.

Overview of Lipid Metabolism01:24

Overview of Lipid Metabolism

902
Lipid metabolism is a crucial process in the human body that involves the synthesis and degradation of lipids. This process is essential for energy production, cell membrane formation, and hormone production, among other functions.
Lipolysis: The Breakdown of Lipids:
Lipolysis is the process of breaking down lipids, particularly triglycerides, into glycerol and fatty acids. This process typically occurs in the adipose tissue and is triggered by various hormones, including glucagon and...

Overview of Metabolism

29.4K
Living cells constantly carry out various chemical reactions which are necessary for their proper functioning. These reactions are interlinked to one another via multiple pathways. The collection of these chemical reactions is known as metabolism.
Plant Metabolism
Sunlight, the primary source of energy in plants, is first absorbed by the chlorophyll pigments present in their leaves. Plants then use this energy to carry out photosynthesis, where water is oxidized into oxygen and carbon dioxide...

Summary

Individuals with phlegm-dampness constitution (PDC) show altered gut bacteria and lower phytosphingosine, increasing metabolic disorder risk. Supplementing with this compound and bacteria can prevent and treat these conditions.

Area of Science:

  • Integrative medicine
  • Gut microbiome research
  • Metabolic disorder mechanisms

Background:

  • Traditional Chinese Medicine (TCM) links phlegm-dampness constitution (PDC) to metabolic disorders.
  • PDC individuals show metabolic gene expression changes, but mechanisms are unclear.
  • The gut microbiota is a potential key to understanding TCM principles.

Purpose of the Study:

  • To investigate the gut microbiota and serum metabolite profiles in individuals with PDC.
  • To elucidate the role of phytosphingosine and Flavonifractor plautii in metabolic disorders within the context of PDC.
  • To explore the therapeutic potential of F. plautii and phytosphingosine in metabolic regulation.

Main Methods:

  • Analysis of gut microbiota and serum metabolites in PDC and control groups.
  • Correlation studies between PDC scores, phytosphingosine levels, and metabolic parameters.
  • In vivo experiments involving fecal microbiota transplantation and supplementation in mouse models.
  • Mechanistic studies on phytosphingosine's interaction with hepatic PPARα.

Main Results:

  • PDC individuals exhibit distinct gut microbiota and serum metabolite profiles, notably decreased phytosphingosine.
  • Flavonifractor plautii synthesizes phytosphingosine, which is negatively correlated with PDC score.
  • Reduced F. plautii and phytosphingosine levels in PDC subjects with normal metabolic indices predict accelerated metabolic disorder development.
  • Supplementation with F. plautii and phytosphingosine ameliorated metabolic disorders via the gut-hepatic axis by activating PPARα.

Conclusions:

  • Decreased F. plautii and phytosphingosine contribute to metabolic disorder susceptibility in PDC individuals.
  • Phytosphingosine directly activates hepatic PPARα, regulating glucose and lipid metabolism.
  • Diagnosing PDC may identify at-risk individuals for early metabolic disorder prevention and advance TCM integration.
