Treatment for Pulmonary Arterial Hypertension: Prostacyclin Receptor Agonists01:23

Treatment for Pulmonary Arterial Hypertension: Prostacyclin Receptor Agonists

139
Prostacyclin receptor agonists are a class of therapeutic agents integral to managing pulmonary arterial hypertension (PAH). These drugs operate by mimicking the action of prostaglandin I2, or PGI2, a naturally occurring compound in the body.
These agonists bind to the IPR receptor situated on the plasma membrane of the pulmonary artery smooth muscle cells. This binding triggers a cascade of reactions known as the GS-AC-cAMP-PKA pathway. This pathway results in the relaxation of smooth muscle...

Drugs for Peptic Ulcer Disease: Prostaglandin Analogs as Mucosal Protective Agents

323
The gastric mucosa produces prostaglandins E2 (PGE2) and prostacyclin (PGI2), crucial in maintaining gastric health. They exert cytoprotective effects, including increasing bicarbonate secretion, releasing protective mucin, reducing gastric acid output, and preventing harmful vasoconstriction. These effects are mediated through various receptors, such as EP1, EP2, EP3, and EP4.
Non-steroidal anti-inflammatory drugs (NSAIDs) can induce peptic ulcers by inhibiting cyclooxygenase, decreasing...

Drugs for Treatment of Constipation-Predominant IBS

124
Pharmacological therapies for IBS-C are designed to alleviate abdominal discomfort and enhance bowel function. In patients with IBS-C, fiber supplements may help soften stools and decrease straining, but may also lead to increased gas production and bloating. Osmotic laxatives like milk of magnesia are frequently used to soften stools and increase stool frequency in IBS-C patients. In addition, two drugs approved for use in severe IBS-C adult cases are linaclotide (Linzess) and lubiprostone...

Antiplatelet Drugs: Prostaglandin Synthesis, P2Y12 and Glycoprotein IIb/IIIa Inhibitors

436
Antiplatelet drugs emerge as frontline defenders against the insidious threat of thromboembolic diseases, where abnormal clots obstruct vital blood vessels. These drugs stand as bulwarks, inhibiting platelet aggregation and clot formation, thereby mitigating the risk of life-threatening conditions like myocardial infarction, coronary artery disease, and thrombotic strokes.
Prostaglandin synthesis inhibitors, exemplified by the widely known aspirin, wield their power by irreversibly acetylating...

IP3/DAG Signaling Pathway01:11

IP3/DAG Signaling Pathway

11.7K
Membrane lipids such as phosphatidylinositol (PI) are precursors for several membrane-bound and soluble second messengers. Specific kinases phosphorylate PI and produce phosphorylated inositol phospholipids. One such inositol phospholipids are the  phosphatidylinositol-4,5 bisphosphate [PI(4,5)P2], present in the inner half of the lipid bilayer. Upon ligand binding, GPCR stimulates Gq proteins to turn on phospholipase Cꞵ. Activated phospholipase Cꞵ cleaves PI(4,5)P2 and...

Adrenergic Receptors: ɑ Subtype01:31

Adrenergic Receptors: ɑ Subtype

1.4K
Adrenoceptors are classified into α and ꞵ classes based on their potencies to catecholamine agonists. α-adrenoceptors show the following order of catecholamine potency:
Adrenaline ≥ Noradrenaline >> Isoprenaline
α-adrenoceptors are further divided into α1 and α2-adrenoceptors.
α1-Adrenoceptors: These receptors are located postsynaptically on the effector organs and cause constriction of smooth muscle mediated by activation of phospholipase...

Summary

Higher levels of 8-iso-prostaglandin-F2α (8-iso-PGF2α), a marker of oxidative stress, are linked to increased cardiovascular risk in hypertensive patients. This biomarker may help refine risk assessment, especially in those with normal kidney function.

Area of Science:

  • Biochemistry
  • Cardiovascular Medicine
  • Clinical Chemistry

Background:

  • 8-iso-prostaglandin-F2α (8-iso-PGF2α) is a key marker of oxidative stress.
  • Elevated 8-iso-PGF2α is implicated in hypertension and cardiovascular disease pathogenesis.
  • Limited data exist on the prognostic value of 8-iso-PGF2α in primary prevention for hypertensive patients.

Purpose of the Study:

  • To investigate the association between plasma 8-iso-PGF2α levels and 10-year cardiovascular risk in hypertensive patients without established cardiovascular disease.
  • To evaluate the correlation of 8-iso-PGF2α with established cardiovascular risk scores (Framingham and ASCVD).

Main Methods:

  • Plasma 8-iso-PGF2α was measured using ELISA in 432 hypertensive patients (aged 40-75).
  • Cardiovascular risk was assessed using Framingham Risk Score (Fr-S) and Atherosclerosis Cardiovascular Disease Risk Score (ASCVD-S).
  • Analysis considered renal function (eGFR) and adjusted for traditional risk factors.

Main Results:

  • Higher cardiovascular risk was significantly associated with elevated 8-iso-PGF2α levels (p < 0.001).
  • 8-iso-PGF2α strongly correlated with Fr-S and ASCVD-S in the overall population and those with normal renal function (p < 0.001).
  • Significant correlations were observed even after adjusting for traditional risk factors, with specific cutoffs identified for high CV risk.

Conclusions:

  • 8-iso-PGF2α shows potential as a biomarker for cardiovascular risk stratification in hypertensive patients.
  • Its utility is particularly noted in patients with preserved renal function.
  • Further research in longitudinal cohorts is recommended to confirm prognostic value and clinical integration for early prevention.
