Abstract:
Palladium-catalyzed carbonylative reactions, used as a tool for the insertion of carbonyl group into organic moieties, is a synthetically useful transformation. Carbonylative Sonogashira coupling is a convenient approach for the synthesis of α,β-acetylenic ketones which are important class of molecules in pharmaceutical and industrial fields. Development of greener approaches and environmentally benign catalytic systems put forward enormous opportunities in this field and moreover, these researches indicate the formation of heterocycles via this carbonylation strategy, which is a major highlight. Herein, we discuss the progress achieved in palladium-catalyzed carbonylative Sonogashira coupling from 2013 to 2024 and its future prospects.

Alkynes to Aldehydes and Ketones: Hydroboration-Oxidation
One of the convenient methods for the preparation of aldehydes and ketones is via hydration of alkynes. Hydroboration-oxidation of alkynes is an indirect hydration reaction in which an alkyne is treated with borane followed by oxidation with alkaline peroxide to form an enol that rapidly converts into an aldehyde or a ketone. Terminal alkynes form aldehydes, whereas internal alkynes give ketones as the final product.

Acid-Catalyzed α-Halogenation of Aldehydes and Ketones
In the first step of the mechanism, the acid protonates the carbonyl oxygen resulting in a resonance-stabilized cation, which subsequently loses an α-hydrogen to form an enol tautomer. The C=C bond in an enol is highly nucleophilic because of the electron-donating nature of the –OH group. Consequently, the double bond attacks an electrophilic halogen to form a...

Acid Halides to Alcohols: Grignard Reaction
Grignard reagents are a source of carbanions and function as nucleophiles. The mechanism begins with the nucleophilic attack by the carbanion at the carbonyl carbon of the acid halide to form a tetrahedral intermediate. Next, the carbonyl group is re-formed, and the halide ion departs,...
