The myocardial infarction-associated transcript (MIAT) promotes atherosclerosis by increasing vascular smooth muscle cell migration, while microRNA-326 (miR-326) inhibits this process. Targeting the MIAT/miR-326 axis offers a potential therapeutic strategy for atherosclerosis.
Area of Science:
- Cardiovascular Biology
- Molecular Medicine
- Atherosclerosis Research
Background:
- Atherosclerosis (AS) is a chronic inflammatory disease characterized by vascular smooth muscle cell (VSMC) migration.
- The role of the myocardial infarction-associated transcript (MIAT) and microRNA-326 (miR-326) in AS progression requires further elucidation.
Purpose of the Study:
- To investigate the functional role of the MIAT/miR-326 axis in regulating VSMC migration during atherosclerosis.
- To explore the potential of this axis as a therapeutic target for AS.
Main Methods:
- Bioinformatic analysis of AS-related datasets (GEO).
- RT-qPCR and Western blot for gene/protein expression analysis in patient samples and VSMCs.
- Transwell and luciferase assays to assess cell migration and molecular interactions.
- RNA-fluorescence in situ hybridization (FISH) for subcellular localization.
Main Results:
- MIAT was upregulated and miR-326 downregulated in AS patients' plasma and ox-LDL-treated VSMCs.
- MIAT expression correlated negatively with miR-326 expression.
- MIAT knockdown and miR-326 upregulation suppressed VSMC migration.
- miR-326 directly targets MIAT, and MIAT promotes VSMC migration by sponging miR-326.
Conclusions:
- The MIAT/miR-326 axis plays a critical role in regulating VSMC migration in atherosclerosis.
- MIAT promotes AS progression by sponging miR-326, leading to increased VSMC migration.
- The MIAT/miR-326 axis represents a promising therapeutic target for AS treatment.
