Background:

  • Cyclin-dependent kinase-like 5 (CDKL5) deficiency disorder (CDD) is a rare X-linked neurodevelopmental disorder.
  • Severe sleep disturbances are a common symptom in CDD patients, but underlying mechanisms are poorly understood.

Purpose of the Study:

  • To investigate the role of CDKL5 in sleep-wake behaviors and EEG activity.
  • To establish a mouse model for studying CDD-related sleep disturbances and developing therapeutic strategies.

Main Methods:

  • Electroencephalogram (EEG) recordings were used to characterize sleep-wake behaviors in male CDKL5 knockout (KO) mice.
  • Conditional knockout (cKO) of CDKL5 in glutamatergic neurons was performed to assess cell-type-specific effects.

Main Results:

  • CDKL5 KO mice exhibited sleep phenotypes mirroring human CDD patients, including sleep initiation/maintenance difficulties and reduced total sleep time.
  • Pre-sleep arousal was observed in KO mice, while circadian rhythm and homeostatic sleep response remained normal.
  • Conditional knockout in glutamatergic neurons also led to reduced sleep time and impaired sleep maintenance.

Conclusions:

  • CDKL5 deficiency directly causes sleep disturbances in CDD.
  • CDKL5 KO mice serve as a valuable model for translational research into CDD sleep treatments.
  • Sleep and EEG parameters may act as biomarkers for CDD clinical trials.
