Immunofluorescence Microscopy01:12

Immunofluorescence Microscopy

11.4K
A fluorescence microscope uses fluorescent chromophores called fluorochromes, which can absorb energy from a light source and then emit this energy as visible light. Fluorochromes include naturally fluorescent substances (such as chlorophylls) and fluorescent stains that are added to the specimen to create contrast. Dyes such as Texas red and FITC are examples of fluorochromes. Other examples include the nucleic acid dyes 4’,6’-diamidino-2-phenylindole (DAPI), and acridine orange.

Total Internal Reflection Fluorescence Microscopy

6.9K
Total internal reflection fluorescence microscopy or TIRF is an advanced microscopic technique used to visualize fluorophores in samples close to a solid surface with a higher refractive index, such as a glass coverslip. TIRF only allows fluorophores in proximity to the solid surface to be excited. When light from a medium with a lower refractive index (such as air) hits the glass coverslip at a critical angle, the light undergoes total internal reflection stead of passing through the glass.

Phase-Contrast Microscopes
In-phase-contrast microscopes, interference between light directly passing through a cell and light refracted by cellular components is used to create high-contrast, high-resolution images without staining. It is the oldest and simplest type of microscope that creates an image by altering the wavelengths of light rays passing through the specimen. Altered wavelength paths are created using an annular stop in the condenser. The annular stop produces a hollow cone of...

Area of Science:

  • Biomedical Imaging
  • Pathology
  • Biochemistry

Background:

  • Immunohistochemistry (IHC) is a standard technique in pathology for visualizing biomarkers in tissues.
  • Conventional IHC relies on labeled antibodies, which can be costly and time-consuming.
  • There is a need for more efficient and accessible methods for biomarker detection.

Purpose of the Study:

  • To develop and validate a label-free IHC imaging strategy.
  • To demonstrate the capability of oblique-incidence reflectivity difference (OIRD) imaging for biomarker mapping.
  • To assess the potential of this method for pathological analysis and drug screening.

Main Methods:

  • Utilized oblique-incidence reflectivity difference (OIRD) imaging technique.
  • Applied OIRD to tissue slices for label-free biomarker visualization.
  • Monitored dynamic antibody binding kinetics.
  • Validated results against traditional fluorescent IHC.

Main Results:

  • Successfully mapped the spatial distribution of biomarkers without labeled antibodies.
  • Demonstrated OIRD imaging's efficacy in label-free IHC.
  • Observed and monitored antibody-tissue binding kinetics.
  • Confirmed the strategy's effectiveness through comparison with fluorescent IHC.

Conclusions:

  • Introduced a pioneering label-free IHC imaging strategy using OIRD.
  • This method obviates the need for labeled antibodies, simplifying the process.
  • The OIRD technique offers a promising tool for pathological analysis, disease diagnosis, and drug screening.
