Abstract:
Cytotoxin (CTX), a three-finger toxin in cobra venoms, is known to cause cytotoxicity, though inconclusive mechanisms are reported. This study discovered the potential cell membrane protein target of CTX using crosslinking differential mass spectrometry. Among 89 proteins identified, annexin A2-CTX was the most stable binding complex with high binding affinity at the nanomolar scale. The findings provide insights into further investigation of the possible internationalisation pathways of CTX through interaction with annexin A2 in mediating cytotoxicity.
Insights
This study identifies annexin A2 as a key cell membrane target for cytotoxin (CTX), a venom component from cobras. This discovery advances understanding of how CTX causes cell damage and death.
Area of Science:
- Biochemistry
- Toxicology
- Cell Biology
Background:
- Cytotoxin (CTX) is a component of cobra venom known to induce cytotoxicity.
- The precise mechanisms by which CTX mediates cytotoxicity remain incompletely understood.
Purpose of the Study:
- To identify the specific cell membrane protein targets of Cytotoxin (CTX).
- To elucidate the molecular interactions between CTX and its potential targets.
Main Methods:
- Crosslinking differential mass spectrometry was employed to identify protein targets.
- Binding affinity of identified complexes was quantified.
Main Results:
- Eighty-nine proteins were identified as potential binding partners for CTX.
- The annexin A2-CTX complex exhibited the highest stability and a high binding affinity at the nanomolar scale.
Conclusions:
- Annexin A2 is identified as a significant cell membrane protein target for Cytotoxin (CTX).
- The interaction between CTX and annexin A2 offers insights into CTX internalization pathways and cytotoxicity mediation.
