Rapid and sensitive quantitation of sesame 2S protein in foods using immunochromatographic analysis

Abstract:
Since allergic reaction triggered by sesame 2S protein can be life-threatening, a sensitive and rapid method to detect sesame 2S protein in foods is urgent needed. In this paper, based on the extraction and purification of sesame 2S protein, monoclonal antibodies against sesame 2S protein were prepared. The optimal antibody pair 7C12C2 was screened according to the performance of lateral flow immunochromatographic analysis (LFIA) strips. The calculated limits of detection (cLODs) of the strips for the detection of sesame 2S protein in milk, biscuits and bread were 14.70 ng/mL, 5.31 and 5.85 μg/kg, respectively. Notably, the results and recovery rates of the strips for spiked samples were in good agreement with those of high-performance liquid chromatography. The testing of positive samples further confirmed the reliability of LFIA strips. Collectively, our proposed strip can be used for the rapid, sensitive and reliable determination of sesame 2S protein in foods.
