Summary

Zika virus (ZIKV) infection in pregnant macaques caused placental damage and fetal infection with both Puerto Rican and Thai strains. This highlights risks of congenital Zika syndrome (CZS) from contemporary ZIKV strains.

Background:

  • Zika virus (ZIKV) infection during pregnancy leads to Congenital Zika Syndrome (CZS), causing severe neurological birth defects.
  • Previous studies demonstrated ZIKV-PR's placental damage and abnormal oxygen transport in a nonhuman primate model.

Purpose of the Study:

  • To investigate placental damage and fetal infection caused by an endemically circulating Thai ZIKV strain (ZIKV-TH) in pregnant rhesus macaques.
  • To compare the effects of ZIKV-TH with the previously studied ZIKV-PR strain.

Main Methods:

  • Pregnant rhesus macaques were infected with either ZIKV-PR or ZIKV-TH at approximately 50 days gestation.
  • Animals were monitored for 40 days post-infection, with analysis of viremia, immune activation, placental architecture, and viral RNA in maternal and fetal tissues.

Main Results:

  • Both ZIKV-PR and ZIKV-TH induced similar courses of viremia and immune activation compared to controls.
  • Both viral strains caused placental architectural changes, including spiral artery remodeling and infarctions.
  • Similar levels of viral RNA were detected in maternal and fetal tissues for both strains.

Conclusions:

  • The Thai ZIKV-TH strain MU1-2017 exhibits similar pathogenic effects to the ZIKV-PR strain in pregnant macaques.
  • This study provides evidence of in-utero infection with a contemporary Thai ZIKV strain, underscoring the ongoing risk of CZS.
