Induced pluripotent stem cells (iPSCs) can generate mesenchymal stem cells (MSCs) via neuromesodermal progenitors (NMPs) in vitro. These iPSC-derived MSCs show adipogenic and osteogenic potential, with their secretome promoting adipogenesis.
Area of Science:
- Stem cell biology
- Regenerative medicine
- Developmental biology
Background:
- Generating unlimited mesenchymal stem cells (MSCs) from induced pluripotent stem cells (iPSCs) is crucial for clinical therapies.
- Identifying specific progenitor populations aids in refining cell differentiation protocols.
Purpose of the Study:
- To generate MSCs from iPSCs via neuromesodermal progenitors (NMPs) using an in vitro protocol.
- To evaluate the osteogenic and adipogenic differentiation potential of these iPSC-derived MSCs (iMSCs).
Main Methods:
- Established in vitro differentiation protocol for iMSC generation via NMPs.
- In vitro cell culture assays, gene, and protein expression analysis for differentiation potential.
- Membrane array and LC-MS/MS analysis for iMSC protein profile and secretome.
Main Results:
- iMSCs demonstrated both osteogenic and adipogenic differentiation capacity in vitro.
- iMSC protein profiles differed from dental pulp and adipose stem cells.
- iMSC secretome significantly promoted adipogenesis in cultured preadipocytes.
Conclusions:
- MSCs with adipogenic and osteogenic potential can be successfully derived from iPSC-derived NMPs in vitro.
- The secretome of iMSCs exhibits adipogenic potential, offering promising therapeutic avenues.
