    Background:

    • The glomerulus plays a critical role in kidney function.

    Purpose of the Study:

    • To confirm the presence of a C3b receptor in the normal human glomerulus.
    • To investigate the presence of this receptor in other species.
    • To evaluate the receptor's status in various kidney diseases characterized by C3 deposition.

    Main Methods:

    • Utilized sheep erythrocytes coated with C3b as an indicator system.
    • Employed fluoresceinated Salmonella typhi coated with C3b.
    • Tested indicator particle binding in human glomeruli and in glomeruli from multiple animal species (mouse, rat, guinea pig, rabbit, rhesus monkey).
    • Examined receptor presence in kidney disease samples with varying C3 deposition patterns (glomerular capillary vs. mesangial) and in glomerular crescents.

    • Confirmed the presence of a receptor for C3b (but not C3d) in normal human glomeruli.
    • Found no detectable C3b receptor in tested animal species.

    • Detected no C3b receptors on proliferating cells within glomerular crescents.

    Conclusions:

    • A specific C3b receptor exists in the human glomerulus, unique among the tested species.
    • The presence and function of this glomerular C3b receptor are altered in certain kidney diseases with glomerular capillary C3 deposition.
    • Further research is needed to elucidate the exact role of this receptor in kidney health and disease pathogenesis.
