Preclinical Development: Overview01:28

Preclinical Development: Overview

4.8K
Preclinical development consists of a series of tests that ensure the safety and efficacy of a new therapeutic compound before it is tested in humans. There are four main phases to this process. First, safety pharmacology tests are conducted to ensure the drug does not produce any acutely harmful effects. These tests examine parameters such as bronchoconstriction, cardiac dysrhythmias, blood pressure changes, and ataxia. Next, preliminary toxicological testing is performed to determine the...

Clinical Trials: Overview

3.4K
Clinical development focuses on how the drug will interact with the human body and encompasses four key phases of clinical trials, each serving a specific purpose in assessing the safety and effectiveness of new drugs. These phases overlap and build upon one another. Phase I involves a small group of healthy volunteers (typically 20-80 individuals) or, in cases where significant toxicity is expected, patients with the targeted disease, such as cancer or AIDS. The volunteers are tested for...

Area of Science:

  • Pharmaceutical Sciences
  • Natural Product Chemistry
  • Sensory Science

Background:

  • Industrial extraction and purification of Cannabis sativa L. are crucial for reproducible therapeutic and sensorial product attributes.
  • Optimizing these processes ensures consistent quality and efficacy in cannabis-based formulations.

Purpose of the Study:

  • To chemically analyze and characterize the sensory attributes of standardized Cannabis sativa L. preparations.
  • To evaluate the effectiveness of deep-cooled ethanol extraction and wiped-film distillation in preserving cannabinoids and terpenes.
  • To assess the impact of purification on the sensory quality and therapeutic potency of Full-spectrum Extract Cannabis Oil (FECO).

Main Methods:

  • Standardized chemotype I Cannabis sativa L. preparations underwent chemical analysis and sensory evaluation.
  • Deep-cooled ethanol was used for selective extraction, followed by wiped-film distillation under deep vacuum (<0.2 mbar) for purification.
  • Cannabinoid and terpene profiles of crude and purified FECO were analyzed at various processing stages.

Main Results:

  • The cannabinoid profile was preserved from the herbal substance to the final preparation, despite decarboxylation.
  • Purified FECO received higher acceptance ratings for color and overall appearance compared to crude FECO, which had a polarizing taste.
  • Terpene markers were remarkably preserved, indicating high profile reproducibility and potential for controlled reintroduction.

Conclusions:

  • The industrial purification process effectively removed non-cannabinoids, enhancing sensory quality while maintaining therapeutic potency.
  • Purified FECO demonstrates improved sensory acceptance and consistent terpene profiles, crucial for product development.
  • Monitoring extraction and purification is essential for optimizing cannabinoid content and sensory characteristics in cannabis preparations.
