Overexpression of R2R3-MYB type transcription factor SpMYB1 enhances cold and drought tolerance in tomato

Abstract:
Drought and cold stress have profound effects on crop growth and development, reducing quality and commercial production. MYB transcription factors play important roles in regulating plant responses to abiotic stresses. However, the specific functions of numerous R2R3-MYB type transcription factors in mediating abiotic stress tolerance remain to be fully elucidated. In the present study, 119 R2R3-MYB family members were identified in Solanum pennellii using the most recent genomic data and performed comprehensive bioinformatics analysis. Further, we found that SpMYB1 exhibited differential expression patterns in various tissues and significantly induced by drought and cold treatments. Meanwhile, overexpression of SpMYB1 gene enhanced drought and cold stress tolerance in transgenic tomato by increasing chlorophyll content, reducing oxidative damage, decreasing membrane lipid peroxidation, and inducing antioxidant activities. Furthermore, the expression of antioxidant and stress related genes (SlDREB2, SlFLW10, SlABF4, SlCAT, SlPOD, SlSOD, SlCAM, SlRubisco, SlMDH, SlGlut, SlABF2, SlABF3) were significantly increased in SpMYB1 OE lines following drought or cold stress. The results show that SpMYB1 is crucial to abiotic stresses, which would be used as a candidate gene for molecular breeding of abiotic stresses resistance in tomato.
