Abstract:
We have developed a readily feasible Mannich-type reaction of alkylazaarenes with aminals, demonstrating excellent functional group tolerance and scalability. Mechanistic studies reveal that the reactivity and selectivity are governed by the electronic effects in conjunction with the stability and steric hindrance of the enamine intermediates. The resulting aminomethylated products hold promising potential for applications in organic synthesis, medicinal chemistry, and ligand design. This protocol offers a versatile tool for the functionalization of azaarene frameworks.
