Odronextamab significantly improved patient-reported outcomes in relapsed/refractory diffuse large B-cell lymphoma (r/r DLBCL). These findings support odronextamab monotherapy as a viable treatment option for r/r DLBCL patients.
Area of Science:
- Hematology
- Oncology
- Clinical Trials
Background:
- Relapsed/refractory diffuse large B-cell lymphoma (r/r DLBCL) presents a significant unmet need.
- Patient-reported outcomes (PROs) are crucial in evaluating treatment efficacy and patient experience.
Purpose of the Study:
- To assess the impact of odronextamab on PROs in patients with r/r DLBCL.
- To evaluate the clinical meaningfulness of observed PRO improvements.
Main Methods:
- The ELM-2 study (NCT03888105) included 141 patients with r/r DLBCL.
- Patients received odronextamab 160 mg weekly after step-up dosing.
- PROs were measured using FACT-Lym, FACT-G, TOI, EORTC QLQ-C30, and EQ-5D-3L.
Main Results:
- Odronextamab significantly improved FACT-Lym Lymphoma Subscale (LymS), FACT-General (FACT-G), Trial Outcome Index (TOI), and Total Score.
- Improvements were also observed in EORTC QLQ-C30 global health status/quality of life (GHS/QoL) and EQ-5D-3L visual analog scale (VAS).
- Improvements in FACT LymS were clinically meaningful, with most patients reporting meaningful improvement or maintenance.
Conclusions:
- Odronextamab monotherapy demonstrated significant improvements in PROs for patients with r/r DLBCL.
- These PRO improvements support the feasibility of odronextamab as a potential treatment option.
- Odronextamab offers a promising therapeutic avenue for patients with relapsed/refractory disease.
