Area of Science:

  • Oncology
  • Molecular Biology
  • Cancer Research

Background:

  • Hepatocellular carcinoma (HCC) is a common cancer with a poor prognosis.
  • Cuproptosis, a novel cell death pathway, shows promise in cancer prognostics, but its role in HCC is unclear.

Purpose of the Study:

  • To establish and validate a prognostic model for HCC based on cuproptosis-related genes (CRGs).
  • To explore the relationship between CRGs, immune infiltration, and underlying molecular mechanisms in HCC.
  • To investigate the functional role of key CRGs in HCC progression.

Main Methods:

  • Identified differentially expressed CRGs and constructed a prognostic model using Cox regression analysis.
  • Evaluated model performance with Kaplan-Meier curves and ROC analysis.
  • Assessed immune infiltration, conducted Gene Set Enrichment Analysis (GSEA), and performed in vitro experiments.

Main Results:

  • Developed a three-gene (AACS, ABCB6, CKAP2) prognostic model for HCC with high predictive accuracy and independence.
  • The model correlated with immune cell infiltration and enriched pathways like cell cycle and DNA replication.
  • In vitro studies confirmed AACS promotes HCC cell proliferation, invasion, and migration.

Conclusions:

  • A validated CRG-based prognostic model for HCC was established, showing significant predictive effectiveness.
  • Findings deepen the understanding of CRGs in HCC and support their use in prognostic prediction.
