Area of Science:

  • Radiation biology
  • Molecular biology
  • Immunology

Background:

  • Assessing DNA damage and cellular response is crucial for understanding radiation effects.
  • Developing efficient protocols for lymphoid tissue analysis is essential for radiosensitivity studies.

Purpose of the Study:

  • To validate a method for isolating mouse lymph node cells for ex vivo irradiation studies.
  • To characterize DNA damage and the expression of key proteins involved in repair, cell cycle arrest, and apoptosis post-irradiation.

Main Methods:

  • Mouse lymph node cell suspensions were subjected to ex vivo X-ray irradiation (2 Gy).
  • DNA damage was quantified using the comet assay.
  • Levels of DNA repair (γH2AX, Rad51, XPA), cell cycle arrest (Cdc25A, p21, p16), and apoptosis (Bax, Bcl-2) proteins were analyzed.

Main Results:

  • Irradiation significantly increased DNA damage (7-fold) and levels of repair proteins (γH2AX, Rad51, XPA) and cell cycle arrest proteins (Cdc25A, p21, p16).
  • Pro-apoptotic protein Bax increased significantly, while Bcl-2 levels remained unchanged.
  • The protocol allows for rapid sample preparation for radiosensitivity assessment.

Conclusions:

  • The developed protocol effectively isolates lymphoid tissue for radiosensitivity analysis.
  • Ex vivo irradiation induces measurable DNA damage and modulates critical cellular response pathways in mouse lymphoid cells.
  • This method is valuable for studying radiosensitivity in various mouse strains and experimental conditions.
