Exploring potential protein-protein interactions driving unusual fatty acid synthesis in pomegranate

Abstract:
Many plant-derived unusual fatty acids (UFAs) possess valuable chemical properties and have potential applications in the food, feed, and oleochemical industries. Despite significant interest, the mechanisms by which plants synthesize and accumulate these structurally distinct fatty acids remain only partially understood. While enzyme substrate specificities involved in UFA-containing storage lipid assembly have been well characterized in many prior studies, the biochemical roles of protein-protein interactions (PPIs) in coordinating UFA biosynthesis have received less attention. Punicic acid (PuA), a conjugated UFA known for its health benefits, is naturally enriched in pomegranate seed lipids. In this study, we investigated the PPIs involved in PuA biosynthesis using Saccharomyces cerevisiae and Nicotiana benthamiana expression systems. Potential protein components that may facilitate PuA enrichment, including conjugase, desaturase, cytochrome b5 (Cb5), hemoglobin, and various transferases, were analyzed. Among eight pomegranate Cb5 isoforms, only selected variants enhanced PuA levels in yeast when coexpressed with the gene encoding the key conjugase. Subsequent in vivo interaction assays revealed protein interactomes that may support PuA synthesis and channeling. Collectively, these findings provide new insights into the biochemistry and molecular organization of specialized lipid metabolism in plants and offer a valuable starting point for future investigations into the physiological roles and production of UFAs.
