Abstract:
Developing economical, high-throughput separation media is critical for efficient antibody downstream manufacturing. In this work, hierarchically gigaporous polystyrene/dextran composite (GPDC) monoliths were synthesized via one-step activator generated by electron transfer for atom transfer radical polymerization (AGET ATRP) using an emulsion templating approach. Poly(1-vinylimidazole) (PVIM) brushes were subsequently grafted onto the GPDC monolith for antibody-selective capture. Herein, the bicontinuous medium internal phase emulsion (MIPE) template was constructed via interfacial self-assembly of dual surfactants at the water/oil interface, i.e., the water-soluble Pluronic F127 (PF127) and the oil soluble amphiphilic diblock glycopolymer (ADG) made in the lab. In addition to functioning as a surfactant, ADG serves as a macroinitiator for ATRP and provides a glycopolymer coating for the hydrophobic PS skeleton. The resulting GPDC monolith exhibited good hydrophilicity and column permeability (5.14×10-12 m2), featuring a hierarchical pore structure with micro- (∼6 nm), meso‑(20-180 nm), and gigapores (180-3000 nm). After grafting PVIM brushes (GPDC@PVIM), the monolith enabled rapid antibody separation from human serum with 98.3 % purity at a flow rate of 2890 cm/h within 3.5 min, and the recovery of hIgG maintained >85 % after 10 consecutive regeneration cycles. This system provides an efficient platform for rapid antibody purification in downstream processing.
