Exploring the involvement of specific gene(s) during pathogen invasion in plants requires explicit strategy or technique(s), which would facilitate in unraveling the functional mechanism of the gene. Here, we have primarily described the process for analyzing the target protein by using immunoblotting approach with an aim to unravel the functional role of protein of interest. Immunoblotting technique where specific antibodies are used ensures the presence or absence of the target protein and also allows confirming the turnover of the protein(s) at different time points of pathogen infection to plants. The plants expressing the target protein is confirmed by the immunoblotting approach, so that they can be used for the plant-pathogen interaction studies. Also, the interacting partners of the target protein are monitored by subsequent immunoprecipitation process with an aim to see the effect of pathogen infection on the protein-protein interaction in Arabidopsis. Essentially, here we focused protein monitoring techniques to investigate the involvement of target protein(s) through ectopic expression in Arabidopsis.
