Background:

  • Developmental dyslexia (DD) is a common neurodevelopmental disorder impacting academic and social skills.
  • While specific brain regions are implicated, large-scale functional coordination deficits in DD are not well understood.

Purpose of the Study:

  • To meta-analyze functional connectivity (FC) studies in developmental dyslexia (DD).
  • To identify consistent and state-specific brain connectivity alterations in individuals with DD.

Main Methods:

  • A meta-analysis of seed-based whole-brain FC studies was performed using AES-SDM.
  • Included 12 task-based studies (226 dyslexics, 232 controls) and 7 resting-state studies (120 dyslexics, 145 controls).

Main Results:

  • Consistently reduced FC between the left inferior frontal gyrus (IFG) and left fusiform gyrus (FFG) in dyslexics across task and resting states.
  • Task-specific findings: hypoconnectivity (left IFG-right cerebellum) and hyperconnectivity (left IFG-bilateral angular gyrus, ACC, left thalamus).
  • Resting-state specific finding: hypoconnectivity (left FFG-posterior cingulate cortex).

Conclusions:

  • DD is characterized by widespread disruptions in brain functional integration.
  • Findings highlight a core neural pathway (left IFG-FFG) and identify potential connectivity-based biomarkers for DD diagnosis.
