Summary

Obsessive-compulsive disorder (OCD) is linked to altered brain network connectivity, particularly in the beta frequency band. These findings suggest electrophysiological biomarkers for OCD diagnosis and targeted treatment strategies.

Background:

Purpose of the Study:

  • To investigate frequency-specific alterations in functional brain connectivity in individuals with OCD.
  • To explore the relationship between neural synchrony, OCD symptoms, and anxiety.
  • To assess the diagnostic potential of electrophysiological network features in OCD.

Main Methods:

  • Resting-state electroencephalography (EEG) data from 88 OCD patients and 100 healthy controls (HCs) were analyzed.
  • Source-space functional connectivity networks were constructed using the weighted phase lag index.
  • Graph-theoretical metrics and machine learning (support vector machines) were employed to analyze network topology and classify groups.

Main Results:

  • OCD patients showed increased beta-band connectivity between the default mode network (DMN) and frontoparietal network (FPN) compared to HCs.
  • Beta-band FPN connectivity correlated with compulsive symptom severity.
  • Reduced global and local efficiency in the beta band were observed in the right visual cortex.
  • Low-frequency (delta and theta) network metrics correlated with anxiety severity.
  • Multi-frequency network features achieved 87.9% accuracy in classifying OCD patients and HCs.

Conclusions:

  • Frequency-specific disruptions in brain networks are evident in OCD.
  • Electrophysiological connectome biomarkers show promise for OCD diagnosis.
  • Findings support the development of frequency-targeted neuromodulation for personalized OCD treatment.
