Abstract:
This review systematically summarizes the various applications of plant-derived natural products in PROTAC technology, specifically as E3 ligase ligands, protein-of-interest ligands, and facilitators for natural product target identification. By harnessing their structural diversity, plant-derived natural products present innovative strategies for addressing undruggable targets and mitigating drug resistance. Additionally, the combination of PROTAC with proteomics offers robust tools for clarifying the mechanisms of action of natural products. Future endeavors aimed at optimizing ligands and linkers, identifying novel E3 ligase ligands, and integrating artificial intelligence with multidisciplinary approaches are poised to generate a new generation of highly effective, low-toxicity therapeutic molecules, thereby catalyzing a paradigm shift in drug discovery.
