Background:

  • Thyroid disease is increasingly recognized for its potential role in neurodegenerative disease pathology.
  • The specific relationship between thyroid dysfunction and amyotrophic lateral sclerosis (ALS) remains incompletely understood.

Purpose of the Study:

  • To investigate the association between thyroid disease, encompassing hypothyroidism and hyperthyroidism, and the subsequent risk of developing ALS.
  • To analyze the impact of thyroid conditions on ALS incidence within a large cohort.

Main Methods:

  • A matched cohort study design was employed using UK Biobank data from participants enrolled between 2006 and 2010, with follow-up extending to 2022.
  • Cox proportional hazards models were utilized to calculate hazard ratios (HRs) and 95% confidence intervals (CIs) for ALS risk.

Main Results:

  • The study included 42,684 individuals with thyroid disease, of whom 80% were female.
  • Thyroid disease showed a moderate association with an increased risk of ALS development (HR: 1.44, 95% CI: 1.02-2.02), consistent after adjusting for childhood height.
  • Hyperthyroidism was potentially associated with higher ALS risk in individuals aged 60 years or younger (HR: 21.22) and in females (HR: 3.02).

Conclusions:

  • Thyroid disease is associated with a moderately elevated risk of developing amyotrophic lateral sclerosis.
  • Further comprehensive research is essential to elucidate the complex interplay between thyroid disorders and ALS pathogenesis.
