Abstract:
Smilax glabra Roxb. (SG) is a functional food and traditional anti-inflammatory remedy, with astilbin as its primary bioactive flavonoid. However, astilbin's sensitivity to pH, heat, and light significantly limits its application in functional food matrices. In this study, we isolated low molecular weight polysaccharides (LP) were extracted from SG and, for the first time, self-assembled with astilbin to form carrier-free nanoparticles without additional excipients. This green, food-compatible approach markedly improved astilbin stability across pH 3-10 and under thermal treatment at 100 °C, maintaining > 91 % retention under stress conditions. NMR analysis indicated strong hydrogen bonding between astilbin's C3' and C4' hydroxy groups and LP's carboxyl groups, likely inhibiting isomerization. FT-IR confirmed chemical interactions, while XRD and DSC revealed that astilbin shifted to an amorphous state in the nanoparticles. Molecular dynamics simulations showed hydrogen bonding and van der Waals forces as the key stabilizing interactions. In vivo studies using a mouse model of atopic dermatitis demonstrated enhanced intestinal absorption and improved anti-inflammatory efficacy of the nanoparticles. These findings highlight a sustainable and excipient-free strategy to improve the physicochemical stability and bioavailability of heat- and pH-labile polyphenols, providing a promising route for the development of functional food and nutraceutical products enriched with Smilax glabra bioactives.
